Single Nucleotide Polymorphisms (SNPs) detection on base sequences of CAD gene in lignin biosynthesis of loblolly pine (Pinus taeda L.).

摘 要:該研究以美國南部重要的造紙工業(yè)用材樹種火炬松為材料,利用Transgenomic公司最近推出的Wave?R DNA片段分析系統(tǒng),快速檢測肉桂醇脫氫酶(CAD)基因在個體間的單核苷酸多態(tài)性,獲得了令人滿意的結(jié)果.首先利用Oligo 5.1軟件對火炬松CAD基因全序列分區(qū)域進行引物設(shè)計,當(dāng)設(shè)定每一區(qū)域能擴增出100~200 bp產(chǎn)物時,共在8個堿基區(qū)域(S1~S8)分別設(shè)計出了最佳正向引物和反向引物;以來自火炬松12株優(yōu)樹自由授粉種子的胚乳中提取的DNA為材料,對CAD基因8個堿基區(qū)域分別進行PCR擴增,有5個堿基區(qū)域(S3,S5,S6,S7,S8)得到了唯一的并且與預(yù)期堿基長度基本一致的PCR擴增產(chǎn)物,對其余3個堿基區(qū)域進一步進行FailSafeTM PCR擴增,它們均在某些PCR緩沖液中得到了唯一的擴增產(chǎn)物;利用WaveR DNA片段分析系統(tǒng),對6個堿基區(qū)域(S2,S3,S5,S6,S7,S8)進行單核苷酸多態(tài)性檢測,結(jié)果表明來自優(yōu)樹2-2的種子與其它11株優(yōu)樹的種子之間存在堿基序列變異,其中在S5,S6區(qū)域檢測到了較大的單核苷酸多態(tài)性,這兩個區(qū)域分別位于CAD基因的第2和第3內(nèi)含子(Intron2,Intron3).
